چکیده

CC chemokine ligand 2 (CCL2) or monocyte chemoattractant protein-1 (MCP-1) is an important chemoattractant for monocytes and macrophages (Deshmane et al, 2009). CCL2 is the main ligand for the chemokine receptor CCR2, which mediates CCL2’s biological activity. CCR2 is a seven transmembrane spanning Gai protein-coupled receptor (GPCR) constitutively expressed at high levels on monocytes and macrophages. CCL2 expression is induced in a variety of tissue resident cells (e.g. endothelial cells) and immune cells (e.g. macrophages) by inflammatory mediators (e.g. TNF, LPS and IFNg). CCL2 mobilizes monocytes out of the bone marrow and recruits circulating monocytes into sites of infection and inflammation. Perhaps more than any other chemokine system, CCL2 and CCR2 have been shown to play important pathological roles in vivo in murine models of disease. For example, in murine models of atherosclerosis, experimental autoimmune encephalomyelitis and obesity-induced diabetes, mice deficient in CCL2 and/or CCR2 have markedly reduced monocyte accumulation in diseased tissue and are protected from disease (Charo & Ransohoff, 2006). Murine studies have also demonstrated

Novel Small Molecules against Two Binding Sites of Wnt2 Protein as potential Drug Candidates for Colorectal Cancer: A Structure Based Virtual Screening Approach

Wnts are the major ligands responsible for activating Wnt signaling pathway through binding to Frizzled proteins (Fzd) as the receptors. Among these ligands, Wnt2 plays the main role in the tumorigenesis of several human cancers especially colorectal cancer (CRC). Therefore, it can be considered as a potential drug target. The aim of this study was to identify potential drug candidates ...

A Novel Combinatorial Approach to Discrete Fracture Network Modeling in Heterogeneous Media

Fractured reservoirs contain about 85 and 90 percent of oil and gas resources respectively in Iran. A comprehensive study and investigation of fractures as the main factor affecting fluid flow or perhaps barrier seems necessary for reservoir development studies. High degrees of heterogeneity and sparseness of data have incapacitated conventional deterministic methods in fracture network modelin...

Comparative Analysis of Expression of Chemokoine Receptors CXCR4, CXCR6, CCR1 and CX3CR in Human Adipose-Drived Mesenchymal Stem Cell with Valproic Acid

Introduction: Chemokine receptors are found on the surface of stem cells. There have been 19 distinct chemokine receptors described in mammals. Chemokines are major players in migration and homing. Therefore, changes in their levels or function can help us to increase the migratory potential of these cells. Valproic acid differs in structure from other drugs in common use. The way in which Va...

A Novel QSAR Model for the Evaluation and Prediction of (E)-N’-Benzylideneisonicotinohydrazide Derivatives as the Potent Anti-mycobacterium Tuberculosis Antibodies Using Genetic Function Approach

Abstract A dataset of (E)-N’-benzylideneisonicotinohydrazide derivatives as a potent anti-mycobacterium tuberculosis has been investigated utilizing Quantitative Structure-Activity Relationship (QSAR) techniques. Genetic Function Algorithm (GFA) and Multiple Linear Regression Analysis (MLRA) were used to select the descriptors and to generate the correlation QSAR models that relate the Mi...

A Novel Highly Potent Therapeutic Antibody Neutralizes Multiple Human Chemokines and Mimics Viral Immune Modulation

Chemokines play a key role in leukocyte recruitment during inflammation and are implicated in the pathogenesis of a number of autoimmune diseases. As such, inhibiting chemokine signaling has been of keen interest for the development of therapeutic agents. This endeavor, however, has been hampered due to complexities in the chemokine system. Many chemokines have been shown to signal through mult...
